Preparation of RV Caps and Grape Seed Extract solution RV Caps solution

2gm content obtained from RV Caps capsules was dissolved in 30ml of dehydrated ethanol. In which 1.88 gm remains in solution leaving 0.12gm as insoluble part. Thus 1.88gm RV Caps content present in 30ml of solution (stock solution).

63µl stock solution (containing 4mg RV caps) was diluted to 500µl by water and fed orally to each mouse (wt of mouse = 20gm) every day.

Thus the dose of RV Caps becomes 200 mg per kg body weight per day

Grape Seed Extract solution

Similarly 4.5gm Grape Seed Extract was dissolved in 30ml of dehydrated ethanol. In which 3 gm remains in solution leaving 1.5gm as insoluble part. Thus 3gm Grape Seed Extract present in 30ml of solution (stock solution).

40µl stock solution (containing 4mg Grape Seed Extract) was diluted to 500µl by water and fed orally to each mouse (wt of mouse = 20gm) every day.

Thus the dose of Grape Seed Extract becomes 200 mg per kg body weight per day

Comparison of the Efficacy of RV caps and Grape Seed Extract

Treatment No. of EAC cells (in millions)

Drug : RV CAPS: Testing of anticancer property of RV CAPS with EAC (Ehrlich Ascites Carcinoma) cells in murine model: Animal models and treatment schedule:

  1. Swiss albino female mice, weight 18-20 gm maintained under standard laboratory conditions.
  2. Contents of the RV CAPS suspended in water which is termed as DRUG.
  3. In each set all the 16 mice were inoculated intraperitoneally with approximately ten lac (1 x 106) EAC ( Ehrlich Ascites Carcinoma) cells in 200 1 of normal saline to each at their peritoneal cavity. These EAC inoculated mice were divided into two groups, (Group I & Group II ) each group containing 8 mice. Day of inoculation with EAC cell is considered as day zero.
  4. One group (Group I ) kept untreated i.e., the mice received orally only 500 1 of water. Another group (Group II) received oral dose of DRUG at an amount of 34 mg/kg body wt/ day i.e., each mouse received 680 g drug in 500 1 suspension everyday orally with help of gastric canula. Treatment started on day 3
  5. On 3rd day immediately before starting treatment two mice (one mouse from each group) were sacrificed and peritoneal EAC cells were counted by Trypan blue dye exclusion method using haemocytometer (result shown in Table 1)
  6. Body weight of each mouse was monitored in regular intervals.
  7. In set 1 experiment on day 11 after receiving 7 doses of drug three mice from each group were sacrificed and EAC cells were counted (by Trypan blue dye exclusion method) from peritoneal lavage (Table 1)
  8. In set 1 experiment on day 16 after receiving 11 doses of drug remaining four mice from each group were sacrificed and EAC cells were counted (by Trypan blue dye exclusion method) from peritoneal lavage (Table 1).
  9. Photograph of the mice in set 1 experiment on day 16 was shown in fig. 1
  10. Similarly in set 2 experiment 16 mice were inoculated intraperitoneally with approximately ten lac (1 x 106) EAC (Ehrlich Ascites Carcinoma) cells in 200 1 of normal saline to each at their peritoneal cavity. These EAC inoculated mice were divided into two groups (Group I &Group II), each group containing 8 mice. Day of inoculation with EAC cell is considered as day zero. On day 15 after receiving 10 doses of drug mice were sacrificed and EAC cells were counted (by Trypan blue dye exclusion method) from peritoneal lavage (Table2)
  11. Photograph of the mice In set 2 experiment on day 15 was shown in fig. 2
  12. Four sets of experiment were done and similar results were found. Two sets of experiments were presented in table I, Fig, 1 and Table 2, Fig. 2 respectively.

Each group consist 5 mice. Each mouse was inoculated with 106 (1 million) EAC cells.

The day of inoculation was considered as day 0.

Treatment with Grape Seed Extract and RV Caps were started from day 3 and continued up to day 13 receiving 9 doses (5doses/week).

Grape Seed Extract, RV Caps were fed orally

Experiment was repeated thrice.

The ascites fluid containing cells were quantitatively removed from the peritoneal cavity on the indicated day and numbers were counted under phase-contrast microscope.

It is evident from the above results that RV Caps is more efficient anti-cancer agent.
